You are only usb 2.0 capable.

Or boot Puppy from a cd, get to the desktop, insert the usb and check
with Gparted your usb.
Select only >>> USB
See if LBA and Boot are ticked for your usb. It was flagged Boot and is now flagged for both.
As I have installed the OS to harddrive, I rebooted, selecting USB where the stick is shown in the Grub bootloader, but an error message gives file not found .
It is recognised and on the desktop when booted from HD and shows all the Puppy files.

You stated your bios only offers these boot device options for USB.
Your only option is to try making it apear as one of these to the bios.

I can live without the facility to boot from stick, so thanks all, but it is not worth your time & effort to pursue this.
