I'm curious what program did you use to burn the CD? I used BURNCDCC here and it worked (both Slacko 5.7 and 6.3.2):

A lot of things that could be affecting booting from a Live Puppy CD.
Maybe some setting in the security section.
Maybe some setting about what device can work or not work
Just look for anything that could affect using a CD or an OS.
The things they do not tell you, are usually the clue to solving the problem.

I would like to revive this thread, since I have the same issue and the OP did not post any solution to it.
I am still a beginner to (Puppy) Linux OS however, in the recent past, I successfuly installed Puppy on two PC's (dual boot) running Windows XP. Lately, I wanted to run Puppy on my desktop and netbook PC's. So I popped the old Live CD into my Dell Optiplex 390, Intel Core i5-2400, 3.10 GHz, 4GB RAM running Win7 64bit and it booted to a point where a message appears: "puppy_slacko_5.7.sfs not found. Dropping out to initial-ramdisk console" That live CD was working fine on the older XP PC's.
Next, I downloaded the new Puppy version slacko-6.3.2-uefi.iso (32bit) und burned the iso to a new DVD using FreeIsoBurner software. No luck, same message. Then I created a bootable USB Flash drive with the same iso using "Unetbootin" software. This worked and I could boot both PC's. So why is the Flash drive booting and not the DVD?
I prefer to boot from the CD/DVD as the Flash drive gets often corrupted. Please help.

Try this.
Copy the puppy_slacko_5.7.sfs and place it in the first partition of the hard drive.
Just in the partition do not place it in any folder
Probably some kind of timing problem with the DVD drive.
Could be how Puppy sees the drive, after the initial boot process starts identifying hardware type.
Just seems like the newer the Drives are the harder it is to get a Live Puppy CD/DVD to boot.
About USB drive corruption.
If you start with a freshly partitioned and formatted drive.
Never let Windows use it for storage.
The drive is probably formatted Fat32. Windows, over time, will fragment the file system, as part of deleting and adding to it.
It should stay uncorrupted with Puppy using it.
If you look in the boot menu entry for Puppy.
Add pfix=fsck to the end of the kernel line.
That will tell Puppy to do a file system check and repair when it boots.

I have only two partitions. The first one is 210GB win7 boot NTFS and the other is 10GB ext3. You mean the ext3 partition?
Also, I compared the files on the DVD and the flash drive. The following files are MISSING on the DVD:
menu.C32
syslinux.cfg
ubnfield.txt
ubnpathl.txt
vesamenu.c32
zdrv_slacko_6.3.2.sfs

So it's hardly surprising you're having problems booting from it.

I think win7 64bit is the culprit. It does not boot the CD-ROM that boots fine in a 32bit XP machine. My plan was to use a Live Puppy CD/DVD to boot and use the ext3 partition for the SaveFile. Oh well, I will use the USB Flash drive for now. I will burn another DVD directly with Win7 file manager and see if it makes any difference. I will be away for a few days and report back upon return. Us a CD/DVD burning program that will run in Windows.
They know how to properly burn an iso image.
Just make sure to select burn iso image.
Several out there you can download for free.
As suggested above.
Even better.
Boot with the usb of Slacko.
Use Pburn to make the live Slacko DVD.
